BACTERIOPHAGE PROJECT
Hi my name is richard I am in sixth year at secondary school and am about to go to university in september to do microbiology. At the moment however I am trying to finsh my advanced higher biology project on bacteriophage. However I am having several problems 1) I am struggling to remove bacteria from the sample even after centrifuging for two minutes and filtering it through a 0.45 micron filter 2) Finding a good source of phage I have collected water from a pond and a sewage outlet pipe and i am getting no plaques forming on ecoli plates. However the left over bacteria from the phage sample could be covering them over again. Any help and general information would be excellent. Thanks in advance. Ricky. |

Make sure that the sewage used is untreated, ask a sewage treatment plant for some if you can. Otherwise try incubating 5ml of your sewage sample with 5ml NA broth inoculated with a coliform(ideally E.coli)overnight. This should produce coliphages and visible plaques at around a 10-8 dilution.
